Axon Guidance: Proteins Turnover in Turning Growth Cones
نویسندگان
چکیده
Accurate navigation by a neuronal growth cone requires the modulation of the growth cone's responsiveness to spatial and temporal changes in expression of guidance cues. These adaptations involve local protein synthesis and turnover in growth cones and distal axons.

Translating Axon Guidance Cues
Recent studies suggest that local translation of mRNA within axons plays a key role in two aspects of growth cone navigation. One line of evidence implies a local synthesis of proteins that mediate growth cone collapse and turning in response to guidance cues; another indicates that growth cones possess the machinery that would allow the local synthesis of receptors for axon guidance cues.
متن کاملNotch Steers Drosophila ISNb Motor Axons by Regulating the Abl Signaling Pathway
The central problem in axon guidance is to understand how guidance signals interact to determine where an axon will grow. Here we investigate a specific axon guidance decision in Drosophila embryos, the sharp inward turn taken by the ISNb motor nerve to approach its muscle targets. We find that this turn requires Notch and its ligand Delta. We show that Delta is expressed on cells adjacent to t...

Understanding how axons are guided to target locations within the brain is of fundamental importance for neuroscience, and is a widely studied area of research. Biologists have an unmet need for reliable and easily accessible methods that generate stable, soluble molecular gradients for the investigation of axon guidance. Here we developed a microfluidic device with contiguous media-filled comp...
متن کاملCalpain-Mediated Proteolysis of Talin and FAK Regulates Adhesion Dynamics Necessary for Axon Guidance.
Guidance of axons to their proper synaptic target sites requires spatially and temporally precise modulation of biochemical signals within growth cones. Ionic calcium (Ca2+) is an essential signal for axon guidance that mediates opposing effects on growth cone motility. The diverse effects of Ca2+ arise from the precise localization of Ca2+ signals into microdomains containing specific Ca2+ eff...
متن کاملHow actin filaments and microtubules steer growth cones to their targets.
Guidance molecules steer growth cones to their targets by attracting or repelling them. Turning in a new direction requires remodeling of the growth cone and bending of the axon. This depends upon reorganization of actin filaments and microtubules, which are the primary cytoskeletal components of growth cones. This article discusses how these cytoskeletal components induce turning. The importan...
